This gel depicts the NEB 1kb ladder and a digestion of the plasmid with the following enzyme(s): NheI and ClaI We expected the following band sizes for this digest: 8.2kb (for linearization alone with NheI); 5.4kb and 2.7kb with double digest. Linearized plasmid is the expected length. ClaI did not cut because the recognition site was Dam methylated.
