A human codon-optimized Streptococcus pyogenes Cas9 nuclease under the control of the CMV promoter. The resulting Cas9 enzyme, when expressed with a gRNA, was shown to efficiently cleave two genomic sites and induce InDels with frequencies of up to 33% in a human cell line.
