- M13 is not lytic; the phage extrude through the cell wall. The replicative RF I form in the cell is double-stranded and circular, and acts like a plasmid. The phage form outside the cell is linear and single-stranded. In vitro DNA synthesis on a cis-DDP-treated template generates a population of DNA fragments, indicating that duplex synthesis has terminated. Cis-DDP forms an adduct with DNA, at positions where multiple 'g' nucleotides occur, that inhibits synthesis in vitro. Trans DDP has the same effect, only on 'gng' sequences, where n can be any intervening nucleotide.
