- Reference: RJ Kaufman et al, Nucleic Acids Research 1991, Vol. 19, No. 16, 4485-4490. pMT21 was derived from pMT2. 76 bp of the 5' UTR of DHFR cDNA was deleted. A XhoI site was inserted to obtain the following sequence immediately upstream of DHFR: 5'-CTGCAG GCGAGCCT GAATTC CTCGAG CCATC ATG-3' (PstI-EcoRI-XhoI). Also, a unique ClaI site was introduced by digestion with EcoRV and XbaI, treatment with Klenow, and ligation to a ClaI linker. This deletes a 250 bp segment from the adenovirus virus associated RNA (VAI) region but does not interfere with VAI RNA gene expression or function.
