Notes
These data and their annotation were supplied to GenBank by Will Gilbert under the auspices of the GenBank Currator Program. Assembled from pIC19H and pEMBL9m by F. Pfeiffer. For construction of pIC7, a synthetic oligonucleotide has been used to replace the pUC8 polylinker and thus to construct a new cloning vector with a different polylinker. The other pIC-vectors are based on this new pIC7 polylinker, which was combined with the existing pUC9 and pUC19 polylinkers in the following arrangements: pIC19 and pICEM19 vectors: pIC19R: EcoRI- Poly (pIC7) -HindIII- Poly (pUC9) -EcoRI pIC19H: HindIII- Poly (pUC9) -EcoRI - Poly (pIC7) -HindIII pIC20-vectors: pIC20R: EcoRI- Poly (pIC7) -HindIII- Poly(pUC19) -EcoRI pIC20H: HindIII- Poly(pUC19) -EcoRI - Poly (pIC7) -HindIII The pICEM minus phasmid vectors were prepared by exchanging a pEMBL8m segment containing the f1 origin against the pUC-derived fragment from the corresponding pIC-vector. The fragment of pEMBL8m used is identical to the corresponding fragment from pEMBL9m. Because pICEM19Hm contains the pUC9 polylinker, pEMBL9m is given in the PARENT section although pEMBL8m has been used in the experiment. Streak to xgal + amp plate and pick a blue colony. Phagemid with polylinker in lacZ' for production of ssDNA. Medium is 1227 LB plus ampicillin. Hosts: E.coli TB1, E.coli. Related vectors: pIC19H, pEMBL8-, pIC7, pUC9, pEMBL9-, pICEM19H+, pICEM19R-.
