Plasmid: lambdaSK17

Plasmid Type
Cloning Method
Restriction digests of the clone give the following sizes (kb): BamHI--25.0, 13.5 (doublet); EcoRI--25.0, 13.5 (doublet); SalI--21.0, 8.8, 8.2, 5.0, 4.4, 3.6. (ATCC staff) Expression vector permitting positive selection (Spi) of recombinants and control of expression using ochre (supB, supC and/or supG, supV) and amber (supE, supF) suppressors. Useful for conversion to phagemid. Permits expression in three reading frames, construction using partial filling-in, and oriented cloning. The sizes of the major components are (kb): arms--20.3, 12.7; stuffer fragment--13.7; phagemid segments--3.9 (longer arm), 0.7. The polylinker sites given are those associated with the longer arm. The sites associated with the other arm are: SalI BamHI EcoRI XhoI NotI NaeI SfiI AvrII NheI NcoI XbaI. The restriction sites to be used for conversion to phagemid are: SalI, ClaI. Cloning capacity: 0.2 - 15.4 kb. The order of the major features in this plasmid is: arm - ClaI - BspMII - SalI - pMB1 ori - ampR - M13 ori - SacI/MCS/ SalI --> - stuffer - SalI/MCS/XbaI/lacZ' - ClaI - SalI - arm.
