- Created by Moore, July 1995, under contract with NCBI. Restriction digests of the clone give the following sizes (kb): EcoRI--8.0; BamHI--8.0; EcoRI/BamHI--4.0 (doublet). (ATCC staff) Expression vector permitting thermal regulation of expression from the PL promoter. [1] Constructed from a BamHI/EcoRI fragment of pBR322 and a 3.96 kb BamHI/EcoRI fragment from bacteriophage lambda, containing N, rexB, rexA, cI and modified by deleting a 711 bp BglII fragment containing cII and portions of the cro and O genes. [1] The order of the major features in the plasmid is: EcoRI - partial O - partial cro - lambda R promoter - lambda M promoter - cIts - rexA - rexB - lambda L promoter - HpaI / N - BamHI - SphI - SalI - pMB1 ori - ampR. [1] The vector contains the following restriction sites (bp from the EcoRI site): BglII--352, 2744; HindIII--871, 996, 1560; PstI--1454, 7190; HpaI--3196; BstXI--3853; BamHI--3956; SphI--4143; SalI--4232; AvaI--5006; PvuII--5647; PvuI--7316.
