- One of a series of lacZY fusion vectors (ATCC 37594-37596) that allow fusions in one of three reading frames and contain flanking DNA for homologous recombination into the E.coli genome. A 1.3 kb HindIII/NruI kanR fragment (with HindIII site filled in) was inserted at the NruI site of a lacZY fusion plasmid. The 6.3 kb lacZY-kanR cassette is released with SmaI. Restriction digests of the clone give the following sizes (kb): EcoRI--10; SmaI--6.0, 3.7; BamHI--10; PstI--5.5, 4.4; HindIII--10. (ATCC staff) Medium is 1227 LB plus ampicillin. Hosts: E.coli.
