- Created by Moore, July 1995, under contract with NCBI. The insert extends from the HaeIII site at 1.4 kb on the map to the AluI site at 2.96 kb. This was constructed in a two-step process. The 5' HaeII site was linkered with EcoRI; the 3' AluI site was linkered with BamHI. The insert contains an internal BamHI site about 0.8 kb from the EcoRI site. The expression of the gene III product facilitates externalization of periplasmic protein by perturbing the permeability of the outer membrane. Medium is 1227 LB plus ampicillin.
