Depositor Comments
PTPalpha(Y789F)-HA was generated using pXJ41PTPalpha as a template with the 5' primer 5'-CGCCAAGCTTGGCCACCATGGATTCCTGGTTCATTCTTGT-3' and the 3' primer 5'-TGTCAAGCTTCATGCGTAGTCTGGCACGTCGTATGGGTACTTGAAGTTGGCAAAATCTGA-3'. The substitution ATA->AAA results in a Tyr789->Phe substitution. HindIII-cleaved PCR products were ligated into pTet-Splice. The plasmid contains a CMV promoter under control of regulatory sequences from the tet operon such that, when cotransfected with the transactivator pTet-tTAk, it expresses protein only in the absence of doxycycline.

Quality Control
Every DNA prep is verified by next-generation sequencing, confirming plasmid identity, sequence integrity, and absence of DNA contaminants. Concentration is confirmed to be within ±2.5% of the stated concentration.
Our production process has been validated to consistently yield endotoxin levels below 1.0 EU/μg. Individual lots are not tested for endotoxin.
