The Multiplex Yeast Toolkit (MYT) is a collection of 96 plasmids which extend the capabilities of the MoClo Yeast Toolkit (YTK, Addgene Kit #1000000061) for genetically engineering the yeast, Saccharomyces cerevisiae.

This kit will be sent as bacterial glycerol stocks in 96-well plate format.

Description

MYT includes a set of new integration vectors and selectable markers usable across common lab strains, as well as additional assembly cassettes to increase the number of transcription units in multigene constructs, CRISPR-Cas9 tools for highly efficient multiplexed editing, and three orthogonal and inducible promoter systems for conditional programming of gene expression. MYT is fully backward compatible with YTK, using the same bottom-up hierarchical assembly of single and multi-gene constructs, based on the Golden Gate cloning technique. Integration vectors and assembly cassettes are pre-assembled and ready to go, straight out of the kit, for convenience. Additionally, all of the alternative cloning methods included in YTK, such as BioBrick compatibility, are also present in MYT, while simplifying yeast in vivo assembly.
