Addgene diagnostic digest

This gel depicts the NEB 1kb ladder and a digestion of the plasmid with the following enzyme(s): XhoI and SpeI.

We expected the following band sizes for this digest: 9.5kb for the single digests (linearization) and 8.4 and 1.1kb for the double digest. We see the expected pattern.

Note that Addgene prepared plasmid from bacteria growing under both Amp only and Amp/Chlor selection. Results from both preps were identical. The attached digest was for the plasmid grown under Amp/Chlor selection.

Addgene Diagnostic Digest

This gel depicts the NEB 1kb ladder and a digestion of the plasmid pLX302 with the following enzymes: BsrGI, NdeI, and NotI-HF. We expect the following:
Digested with BsrGI--3 bands: 7.9, 1.3, and 0.4 kb.
Digested with NdeI--1 band: 9.6 kb.
Digested with NotI-HF--2 bands: 6.7 and 2.8 kb.

We see the expected pattern
