DNA prep
There was some concern that since this plasmid was so large, it should not be stored or distributed in bacteria. Addgene successfully prepped DNA from our bacterial stock using a the low copy protocol from a Qiagen miniprep kit. The only change in the protocol was that the culture was grown for 22 hours instead of overnight.
This gel also depicts an EcoRI digest of the plasmid. Note that since we do not have full sequence for this plasmid, we could not predict the digest pattern.
