Derived from pCvGAPDH-NanoLuc^opt, a pBluescript-based vector containing the Chromera velia GAPDH promoter (1090 bp; VEuPathDB: Cvel_5775), the first intron of C. velia β-tubulin (106 bp; VEuPathDB: Cvel_33153), the GAPDH Kozak sequence, a NanoLuc CDS codon-optimized for C. velia, and CvGAPDH 3'UTR (632 bp). Backbone was generated by digestion with PstI, which cuts immediately upstream of the Kozak sequence, and AflII, which cuts immediately downstream of the NanoLuc stop codon.
A gBlock fragment that includes the Kozak sequence, CDS of Chromera velia histone H2B (351 bp; VEupathDB: Cvel_19200) including the NLS, a 13-aa linker, and mStayGold codon-optimized for Chromera (675 bp), with both ends containing ~20-bp sequence that overlaps with the backbone insertion sites was synthesized (IDT Inc.) and inserted into the PstI and AflII sites of pCvGAPDH-NanoLuc^opt using Gibson assembly.
