Modifications to backboneAn AscI and an FseI were introduced into the multiple cloning site. The CD45 minigene with the C77G mutation is inserted between the Asc1 and FseI sites. Two LoxP sequences were introduced, one between the Asc1 site and the start of the minigene and another at the end of the minigene in front of the FseI site.

MutationC77G mutation (cysteine to glycine at amino acid position 77) in exon 4 that disrupts normal splicing of this exon. A single C to G nucleotide exchange in exon 4 was introduced into pVGI-1084 to generate C77G CD45.1 expression.
