Human Interferon-Stimulated Gene CRISPR Knockout Library
(Pooled Library #125753)
This CRISPR knockout pooled library targets a set of human interferon-stimulated genes (ISGs). There are 8 guides per target gene, primarily selected from existing libraries (Brunello, GeCKOv2, TKO Version 1 and Human Activity-Optimized CRISPR Knockout Library).

This library is delivered as suspended DNA in a microcentrifuge tube on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze-thaws.

This Human Interferon-Stimulated Gene library was developed in collaboration with the Overbaugh and Paddison labs at Fred Hutch.
This ISG-targeting CRISPR KO library was used in Roesch et al (2018) to screen for ISGs that inhibit VSV/G-pseudotyped lentiviral particles. The curation of the list of ISGs and the library design and synthesis are described in Ohainle et al (2018). The same pooled sgRNAs have also been cloned into an HIV-specific CRISPR knockout plasmid, the HIV-CRISPR vector, to create an HIV-specific CRISPR KO library, called the PIKA-HIV (Packageable ISG Knockout Assembly). This CRISPR KO library specific for HIV research is available through the NIH AIDS Reagent Program (Catalog #13566).
The depositing lab recommends using MAGeCK algorithm for analysis.
These pooled libraries were created by your colleagues. Please acknowledge the Principal Investigator, cite the article in which the plasmids were described, and include Addgene in the Materials and Methods of your future publications.
