STICR Pooled Libraries
(Pooled Library #180483, #186334, #186335)

  • Purpose

    The "single-cell-RNA-sequencing-compatible tracer for identifying clonal relationships" (STICR) barcoding libraries are lentiviral vectors encoding a CAG-GFP transgene with a pre-defined, error-correctable molecular barcode within the 3' UTR. This barcode can be captured by oligo(dT)-based scRNA-seq platforms, including the 10X genomics scRNA-seq 3' platform. See schematic of barcode structure below.

    Each STICR library comprises 50–70 million unique sequences, allowing labeling of a starting cell population and clonal lineage/fate tracing. In addition, each individual library contains a unique constant "index" region within the barcode. This can be used to distinguish cells infected with different libraries.

    Index Sequences

    • Index1: ATC
    • Index3: GAG
    • IndexE: No additional sequence. It’s “index” is the absence of additional sequence.

    Note: Index2 library was designed, but not created. Index numbering was kept the same for consistency in our record keeping.

This library is delivered as suspended DNA in a microcentrifuge tube on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze/thaws.
