Shuttle reporter plasmid to test expression of bleomycin resistance in transformants. Expresses the mutant beta-tubulin gene conferring benomyl resistance. A 2.5 kb SalI/HindIII fragment contains the complete tubulin gene and 5' and 3' flanking regions. The order of the major features in this plasmid is:pUC8 - EcoRI - BamHI - am promoter - ble - XhoI - am 3' region - tub-2 (benomyl resistance) - pUC8. Restriction digests of the clone give the following sizes (kb): BamHI--5.1, 2.1; HindIII--4.0, 3.2; BamHI/HindIII--2.6, 2.5, 1.5, 0.65; BamHI/XhoI--3.0, 1.35, 1.2, 0.9, 0.8; SalI/HindIII--3.3, 2.6, 0.85, 0.55. (ATCC staff) A (GT)n repeat. (personal communication) Medium is 1227 LB plus ampicillin. Hosts: E.coli HB101, Aspergillus nidulans, Neurospora crassa, E.coli.
