Reference: RJ Kaufman et al, Nucleic Acids Research 1991, Vol. 19, No. 16, 4485-4490. Elements: SV40, MLP, TPL, IVS, EMC-L, DHFL, SV40-pA, VA. pMT2-ECAT1 was digested with EcoRI and PstI resulting in a 2752 bp fragment. This fragment was digested with TaqI yielding an EcoRI-TaqI fragment of 508 bp. A 68-bp adapter sequence was synthesized with a 5' TaqI and a 3' XhoI end. This sequence matches the EMC virus leader sequence from nt 763-827. It also changes the ATG at position 10 within the EMC virus leader to an ATT. A three way ligation of the pMT21 EcoRI-XhoI fragment, the EMC virus EcoRI-Taq1 fragment, and the 68 bp oligonucleotide TaqI-XhoI adapter resulted in pED4.
