- pFL39 was constructed from pUC19 plasmid where two alu I sites were modified. The site 629 was replaced by a BglII linker and the site 747 by a ClaI site. The yeast selectable marker has been cloned in the BglII site and the yeast replication origin in the ClaI site. The order of the major features in this plasmid is:EcoRI/MCS/HindIII - TRP1 - ARS-CEN6 - pMB1 ori - ampR. YC-type low copy number shuttle vector permitting visual detection of recombinants based on alpha complementation. The loss per generation (percentage having lost all the plasmids after 20 h of non-selective growth) is 11.0. Restriction digests of the clone give the following sizes (kb): BamHI--4.6; BglII--3.7, 0.85; ClaI--3.75, 0.8. (ATCC staff) Medium is 1227 LB plus ampicillin.
