- The BamHI site was removed from trpC by sodium bisulfite mutagenesis. A 2.1 kb HindIII/SalI fragment (pJB8) containing lambda cos was ligated to HindIII/SalI-digested pBR329. A 4.1 kb XhoI fragment with wild-type Aspergillus nidulans trpC was ligated into the SalI, and a BamHI linker into the NruI site of pBR329. This is a cosmid vector for isolating genes by complementation of mutations in Aspergillus nidulans. It can also be used for library construction. Medium is 1227 LB plus ampicillin.
