Laker et al. Proc Natl Acad Sci USA Vol 84, p 8458. M5-neo was constructed by subcloning several fragments derived from the original MPSV-containing plasmid and the Tn5 gene and subsequent ligation by way of polylinkers. Briefly, a PstI fragment containing the 5'LTR and 5' splice signal, a HindIII-SmaI fragment containing the 3' splice signal, and a HindIII-BglII fragment containing the 3'LTR from MPSV, as well as a SmaI-SalI fragment with the neo resistance coding region of Tn5, were ligated in the BglII-HindIII site of pBR322 (minus EcoRI site).
