- Restriction digests of the clone give the following sizes (kb): BamHI--21.0; EcoRI--21.0; HindIII--6.8 (doublet), 3.6, 1.45, 1.1. (ATCC staff) pBR322 cut with BamHI, and pMS1 (a 19 kb plasmid from Rhodococcus equi) partially digested with Sau3AI, were ligated to form pMS2. The construct retains ampR and ori from pBR322 and kanR and ori from pMS1. A shuttle vector stably maintained in E.coli as well as members of the coryneform group, namely Corynebacterium ATCC 13059 and Rhodococcus equi. Ampicillin and kanamycin resistance are expressed in E.coli; kanamycin resistance is expressed in Rhodococcus equi. Medium is 1227 LB plus ampicillin.
