- A 480 bp HindIII/Sau3AI fragment from pTiA6 was isolated, the Sau3AI site changed to BamHI, and the ends filled. HindIII linkers were added and it was cloned into the HindIII site of MTX4 (ATCC 39276). An episomal Saccharomyces cerevisiae/E.coli shuttle vector. Restriction digests of the clone give the following sizes (kb): HindIII--12.5; PvuII--10.5, 2.45; BamHI--12.5. (ATCC staff) Medium is 1227 LB plus ampicillin.
