- Plasmid pTY24 is built from pFA6aGFP-KanMX6 (See Wach et al.) as the parent vector. pDsRed.T1.N1 (see Bevis & Glick) was used as a template to amplify by PCR a fragment containing DsRed. The forward PCR primer introduced a Pac1 site, and the reverse primer introduced an Asc1 site. The PCR product was then introduced into the unique Pac1 and Asc1 sites of pFA6a... plasmid, replacing GFP.
