Created by Moore, July 1995, under contract with NCBI. The sequence and reading frame of the multiple cloning sequence is: 5' G AAT TCC CGG GGA TCC GTC GAC CTG CAG CCA AGC TTC GAT CCC 3'. The sequence begins with the first nucleotide of the MCS & ends with the CCC for amino acid 8 of beta-galactosidase. One of 3 promoter-cloning, YE type shuttle vectors (ATCC 37725 to ATCC 37727) with URA3 selection in Saccharomyces cerevisiae, a beta-galactosidase reporter gene and multiple cloning sites differing in reading frame. The cleavage position in the reading frame for cloning sites is (where 3 = between triplets): EcoRI-3; SmaI-1; BamHI-1; SalI-1; PstI-2; HindIII-3. Restriction digests of the clone give the following sizes (kb): EcoRI--8.0; HindIII--8.0; SmaI--8.0; BamHI--8.0; PstI--8.0. (ATCC staff) Medium is 1227 LB plus ampicillin.
