pUC18 was digested by HindIII and EcoRI, then DNA fragment containing maize ubiquitin promoter, E. coli uidA gene(GUS) and NOS terminator which is obtained from pAHC25 was inserted. GUS region was cut out by using SmaI and SacI, and Gateway cassette C1 which is obtained from pMDC32 was inserted there. SmaI site was destroyed by this procedure.
