Purpose
The Cultivarium MACKEREL (Modular, NGS-trACKable ExpRession Element) Library is a library of broad host range prokaryotic promoter-RBS regulatory sequences formatted as a golden gate assembly Level 0 part. The library consists of 421 prokaryotic promoter-RBS sequences mined from mobile genetic elements and shown to drive transcription at a range of strengths across different species of bacteria in cell-free systems by Yim et al. (2019). Since this library adopts a widely used golden gate assembly syntax, it is cross-compatible with a number of golden gate toolkits available through Addgene (GoldenBraid, MoClo, CIDAR, POSSUM and more).
