A RBP PCR product from genomic E. coli DNA encoding mature RBP was cloned into the KpnI site of FLIPglu-600u in pRSETB, replacing the glucose sensor's mglB moiety. The RBP sequence was found to carry a D208G substitution, which does not significantly affect the binding affinity.
