Modifications to backboneA fragment containing two BbsI sites for Golden Gate assembly of the 20 bp crRNA oligonucleotide was inserted upstream of the gRNA scaffold sequence by Gibson assembly. The kanamycin resistance cassette was replaced with a spectinomycin resistance cassette. A BbsI site in the plasmid backbone was removed.
