Depositor Comments

Authors were not aware of the 493 base pair deletion in the backbone, which includes a part of ITR. They were able to package the plasmid into AAV1/2 capsids, however, since mutations of ITRs can lead to reduced efficiency in viral packaging, they recommend to reclone the construct into an AAV vector with intact ITRs.

The AAV backbone containing the 1.1-kb CMV enhancer/chicken h-actin (CBA) promoter, the woodchuck posttranscriptional regulatory element (WPRE), and the bovine growth hormone polyA was derived from a plasmid made by Matthias Klugmann (Mol. Cell. Neurosci. 2005, 28:343-360 https://doi.org/10.1016/j.mcn.2004.10.002)
