The forward oligo included SapI restriction sites, the duplicated coat protein promoter sequence, and 20-bp sgRNA sequences flanked by the 5' sequences of the sgRNA scaffold. A common reverse oligo (esgRNA in FoMV R) was used to amplify the 3' region of the sgRNA scaffold, including a SapI restriction site for subsequent cloning.
