Depositor Comments
Intermediate vector, referred to as a gRNA insert vector, is used to clone guide RNA sequences by annealing complementary oligonucleotides with appropriate overhangs and inserting them into a BbsI site positioned between the PpU6 promoter and the gRNA scaffold. The PpU6 promoter and gRNA scaffold are flanked by outward-facing BsaI sites with unique four-base overhangs, enabling directional and ordered assembly via Golden Gate.
