Usage/Comments: This plasmid expresses ΦC31 integrase and is used for marker curing and plasmid backbone excision after payload integration. When payload plasmids are designed with ΦC31 att sites flanking the selectable marker and backbone, introduction of this plasmid by conjugation catalyzes recombination between att sites, excising the antibiotic resistance cassette and vector backbone. This enables generation of markerless strains suitable for downstream engineering cycles or deployment. Counterselection (e.g., sacB-based sucrose selection) may be used depending on the payload design. Please visit https://doi.org/10.64898/2026.04.17.717921 for bioRxiv preprint.
