- There is no XbaI site even though restriction site cluster indicates one. [2] This is a plasmid vector that maintains 20 copies/cell at 30C but replicates in an uncontrolled manner at 37C until cell death occurs (doubling copy number every 15 min.). [2] Restriction digests of the clone give the following sizes (kb): EcoRI--4.3. (ATCC staff) The vector was contructed from the following sequences (EcoRI site as nucleotide 1):0-35, multiple cloning site; 37-235, terminator; 416-1259, copA, copB; 1260-2879, repA, ori; 2880-4008, bla; 4089-4465, stability sequence. [2] The sequence from nucleotides 4089 to 4465 were derived from pSC101 (ATCC 3702) and can be removed from this construct by an EcoRI/SstI double digest. [2] Medium is 1227 LB plus ampicillin.
