- In the absence of the products of nifA, ntrA, or both the transcription start site of the nifH/lacZ gene, with a point mutation in the promoter region, is shifted from the wild-type nifH mRNA start site at position 101 to positions 60-62. However, when the products of nifA and ntrA are both present transcription starting at positions 60-62 is abolished and the normal transcription start site at position 101 is used. In wild type plasmid the upstream start sites at 60-62 are also used in the absence of the nifA and ntrA products.
