- Created by Moore, July 1995, under contract with NCBI. Contains the lac operator. The PvuII site is located 5 bp downstream of the lacZ ribosome binding sequence. [2] The tac promoter can be excised by a EcoRI/PvuII digest. [2] A 55 bp HpaII/PvuII fragment containing the -10 region of the lacUV5 promoter was inserted into the ClaI site of pEA300 (ATCC 37181).
