- YC-type shuttle plasmid which can be used as a reporter of chromosome segregation due to loss of SUP11-containing chromosomes. Constructed by inserting a 0.8 kb EcoRI/HindIII fragment containing ARS1 and a 6.0 kb BamHI/SalI fragment containing CEN4 into the corresponding sites of YRp14. Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--6.1, 2.3, 1.6, 0.8, 0.7; BamHI--11.5; EcoRI--5.5, 4.5, 1.5; SalI--11.5. (ATCC staff) The order of the major features in this plasmid is: ARS1 - BamHI - CEN4 - SalI - URA3 - SUP11 - rep - pMB1 ori - bla. Medium is 1227 LB plus ampicillin.
