- A 570 bp SalI/EcoRI fragment containing the PHO5 promoter was ligated to a 5965 bp BamHI/SalI fragment of YRp7-32cat (ATCC 37624). The depositor recommends using YRp7-17camp (ATCC 37630) as the reference for CAT expression and using the CAT assay methodology of Neumann, et al. One of a series (ATCC 37624 through 37634) of Escherichia coli/Saccharomyces cerevisiae shuttle vectors for investigating control of expression in yeast. Restriction digests of the clone give the following sizes (kb): EcoRI--4.5, 1.37, 0.55; PstI--5.5, 1.3; PvuII--4.7, 2.15. (ATCC staff) Medium is 1227 LB plus ampicillin.
