Purpose
TCR Rapid Assembly for Functional Testing (TCRAFT) enables efficient assembly of paired αβ T-cell receptor (TCR) libraries from sequences by accurately linking TCR variable region genes to CDR3α/β-containing oligos through three Golden Gate reactions. The TCRAFT kit consists of four vector pools (1A, 1B, 2A, and 2B) encoding TCR components and a destination expression vector, pHIV-LacZ-SapI (Plasmid #237610).
To build a TCR library, users generate two oligo pools with the TCRAFT script linked in the protocol and perform three simple Golden Gate assembly reactions with the four vector pools in this kit. The resulting TCR library can be expressed in cell lines or primary cells for functional screening.
