Modifications to backbonePlasmid assembled from 3 CYANO-VECTOR modular devices: pCVD021 / CV-S7942NS2: The device comprises the chromosome integration site NSII for Synechococcus elongatus PCC7942. The device has an origin of replication for E. coli and a bom site for conjugal transfer, which are derived from pBR322. pCVD003 / CV-aph1: The device confers kanamycin or neomycin (Km/Nm) resistance. pCVD015 / CV-ccdB-SwaI: The device comprises the ccdB gene (toxic for commonly used E. coli cell line), which can be taken out with the SwaI endonuclease and swapped for the sequence(s) of interest using adaptors optimized for seamless assembly reaction.
