We extensively modified the plasmid of pLVX-AcGFP-C1 (Clontech, 632155) by replacing CMV promoter with EF1a promoter, adding P2A to mediate co-expression of the gene of interest, and removal of PGK promoter and puromycin resistant gene to make a lentiviral backbone vector of pLVH-EF1a-P2A.
