Purpose
These pooled CRISPR knockout libraries target RNA-binding proteins (RBPs). Pooled library #141438 targets 1,078 RBPs using 10 guide RNAs per gene. Pooled library #250988 targets 206 RBPs with 5 single-guide RNAs (sgRNAs) per gene in the lentiCRISPR v2 backbone, enabling doxycycline-inducible expression of Cas9 after establishment of library-expressing cells in vivo.
Library #250988 was prioritized from previous CRISPR screens that identified RBPs essential for survival in KRAS-, BRCA1-, and MYC-driven cancers, making it relevant for probing cancer-specific vulnerabilities. Its reduced size, combined with the doxycycline-inducible design of the lentiCRISPR v2 backbone, makes it particularly well-suited for in vivo screening such as within a tumor xenograft context.
