  • Purpose

    Leishmania mexicana Cytosine Base Editing v1 (LmexBE_V1) is a genome-wide loss-of-function CRISPR base editing library designed to introduce STOP codons within the first 50% of coding sequences in the genome of L. mexicana. Libraries are transfected using the technique described in Herrmann-May, 2025 (Link opens in a new window). The library can be used for a range of screens in L. mexicana, such as fitness and drug resistance screens.

  • A stable cell line is required for the screen, expressing a Cas9 base editor, a Cas12a nuclease, and a T7 RNA Polymerase. Can be used in conjunction with either pTB106 or pTB107, they differ only in the type of base editor that is expressed. Recommended plasmids that allow for the expression of the required proteins include pTB106-Hyg (Addgene #236779), pTB106-Phleo (Addgene #236780), pTB107-Hyg (Addgene #236781), or pTB107-Phleo (Addgene #236782).
