pMVP (plasmid-based modular vector platform) and pMAGIC (plasmid-based modular adenovirus for genomic interrogation with Cas9) are innovative and versatile cloning systems that allow for the rapid generation of unique adenoviral, lentiviral, expression, PiggyBac transposon, and Sleeping Beauty transposon vectors for transgene or RNAi delivery and dCas9-based epigenetic engineering.

Description

pMVP (plasmid-based modular vector platform) uses MultiSite Gateway cloning and enables the rapid creation of unique vectors containing a gene of interest in the context of an array of promoters, conditional expression modalities, fluorescent reporters, and molecular tools. These components can be recombined into 35 destination vectors allowing for the generation of expression, Sleeping Beauty transposon, PiggyBac transposon, adenoviral, and lentiviral vectors. In total, pMVP permits a gene of interest to be incorporated into >108,000 unique vector permutations that are exquisitely tailored to match experimental goals.

pMAGIC (plasmid-based modular adenovirus for genomic interrogation with Cas9) builds upon pMVP to allow simple and rapid assembly of complex SaCas9 and xCas9(3.7) gene- and epigenome-editing vectors. The compatibility of pMAGIC with adenovirus vectors enables efficient delivery of multiple gRNAs alongside Cas9 or dCas9 fusions with VPR, KRAB, p300core, LSD1, or DNMT3a-3L into primary cells.
