Purpose
The Autodisplay Toolbox (ATB) is a collection of plasmids (pATB) that enable the display of recombinant proteins at the surface of Gram-negative bacterial cells (autodisplay). Based on a classical (EhaA) and an inverse (YeeJ) autotransporter protein, variants were constructed with different signal peptides, linkers, and adjacent regions. To account for different expression needs, gene expression was set under control of three different promoters (ParaBAD, PrhaBAD, and PRox306) resulting in three separate plasmid libraries (one library per promoter, 27 plasmids per library, 81 pATB in total). The pooled plasmids contain a passenger domain to be cloned out and replaced with the insert of interest. The ATB can then be used to screen for optimized bacterial surface display for a protein of interest.
