CROPseq-iT7 Human CRISPR Knockout Pooled Library (Brunello)

  • Purpose

    This library combines gRNAs from the Brunello Human CRISPR Knockout Pooled Library (Addgene #73178) with an inverted T7 promoter downstream of the guide cassette to enable genome-scale optical perturbation screening. This method uses fluorescence microscopy for optical phenotyping in living cells and Nuclear In-Situ Sequencing (NIS-Seq) to identify individual gRNAs in the same cells. In principle, any phenotype observable by microscopy and any nucleated cell type is thus compatible with CRISPR perturbation screening.
