Deposited by Christopher Voigt's lab, these response function plasmids (pRF-) contain a transcriptional repressor, which controls the expression of a YFP output. Repressors are under the control of the IPTG-inducible Ptac promoter. Expression of the YFP gene is controlled by a synthetic constitutive promoter that contains a repressor operator sequence. The YFP output is repressed in the presence of IPTG. The various circuit plasmids achieve either the NAND (pLux-LmrA and pNAND-PhlF) or AND (pAND) behavior. To generate the NAND behavior, the following input concentrations were used: no inducer (-/-), 1 mM IPTG (+/-), 20 µM 3OC6HSL (-/+), and 1 mM IPTG and 20 µM 3OC6HSL (+/+).To generate the AND behavior, the following inducer concentrations were used: no inducer (-/-), 1 mM IPTG (+/-), 100 ng/mL aTc (-/+), and 1 mM and 100 ng/mL aTc (+/+).
