Description

This collection contains a set of 120 lentiviral vector plasmids. Each plasmid encodes a unique protein barcode (Pro-Code), which can be identified on cells by staining with a cocktail of just 10 antibodies. Each Pro-Code is comprised of a mutant version of the human NGFR gene (dNGFR) fused to a sequence encoding three different linear epitopes. The Pro-Code is expressed from the human EF1alpha promoter. The lentiviral vector also contains the U6 promoter and a unique cloning site for insertion of a CRISPR guide RNA. The Pro-Code vectors can be used to carry out pooled CRISPR screens in which 120 different CRISPR-carrying cell populations can be resolved by staining cells with a 10 antibody cocktail and performing single cell flow cytometry, mass cytometry, or by multiplex imaging. The Pro-Code vectors can also be used for tracking cell clones or cell populations.
